Description
The clinical significance of the loss of the Y chromosome in bone marrow specimens has been under debate since the finding was first observed in 1966. Whether loss of the Y chromosome represents only a normal aging phenomenon, as opposed to representing an abnormal clone associated with myelodysplastic syndrome (MDS) and/or acute myelogenous leukemia (AML), remains difficult to evaluate because of the increasing incidence of MDS and AML with age. Multiple studies have attested to loss of the Y chromosome as part of the normal aging process. (2-4) However, loss of the Y chromosome also has been suggested to represent a leukemic clonal phenomenon as evidenced by the return to a normal karyotype during remission from acute leukemia. (5,6) In 2000, a study of 215 male patients showed that loss of the Y chromosome in more than 75% of cytogenetically analyzed cells was associated with a hematologic disease process. (7) This study attempts to evaluate the significance of varying percent ages of Y chromosome loss as it relates to AML or MDS, with adjustment for age. MATERIALS AND METHODS
